The Faculty of Health and Medical Sciences (SUND) and Novo Nordisk A/S established a research centre in 2010 - The LIFEPHARM Centre ( www.lifepharm.dk ) - with a total budget of DKK 78 million covering activities until the end of 2020.
The present position is part of the Centre´s activities. The purpose of the research centre is to strengthen the research in experimental pharmacology and toxicology and to increase the output of candidates and PhDs with a strong in vivo pharmacological profile.
It is expected that the future PhD fellow will be enrolled in the Graduate Programme for In Vivo Pharmacology and Experimental Animals (IVP) at SUND and participate actively in this network. The IVP Graduate Programme was established in 2006 as a collaboration between the Faculties of Life Sciences, Pharmaceutical Sciences and Health Sciences at the University of Copenhagen, University of Aarhus, and industrial partners; Novo Nordisk A/S, LEO Pharma and H. Haemophilia A (HA) is a chronic bleeding disorder that requires life-long treatment. Current treatment options include recombinant or plasma derived human factor VIII (rFVIII). The major difficulties for efficient long-term treatment are anti-drug (i.e. anti-FVIII) antibody (ADA) development, which occurs in about 30% of patients 1 , the necessity of frequent i.v. infusions and the development of bleeding associated complications like knee and ankle arthropathy 2 . There is currently no appropriate model to investigate and characterise novel compounds for prophylactic treatment in HA. Moreover, the pathological role of immune cell subset in haemophilic arthropathy has not been thoroughly described.
The present PhD project aims to evaluate different methodologies for chronic treatment in pre-clinical HA models, using either immunomodulators or B and T cell deficient mice (Rag2-KO). Parameters such as bleeding phenotype, ADA development, PK and PD will be described. When a methodology for chronic studies has been established, immune cell subset will be either depleted or engrafted in F8-KORag2-KO mice with induced haemophilic knee arthropathy. In this setup the development and progression of haemophilic knee arthropathy will be monitored by histopathology, IHC and potentially µCT. Using these approaches, it will for the first time be possible to study the prophylactic potential of novel compounds for more than two weeks, and further it will be possible to dissect the immunopathology involved in haemophilic knee arthropathy in F8-KO mice.

Terms of employment
Salary and other terms and conditions of appointment are set in accordance with the Agreement between the Ministry of Finance and AC (Danish Confederation of Professional Associations) or other relevant professional organisations. Depending on seniority, the monthly salary begins around DKK 25.141 (ca. 3.374 EUR) plus pension. The position is covered by the Job Structure for Academic Staff at Universities (2013).
